摘要:考察了大孔吸附树脂对柱状假丝酵母脂肪酶 ( CCL)的固定化效果。实验证明大孔吸附树脂 HZ- 841作为固定化载体可以为酶营造一个良好的微环境 ,对水分具有较强的缓冲能力。当固定化酶的含水量为 8%时催化效率最高。醇既是底物又是竞争性抑制剂 ,它的浓度对反应具有较大的影响 ,在萘普生浓度为 2 0 mmol/L时正辛醇最适浓度为 40 mmol/L。选择弱极性大孔吸附树脂HZ- 841固定化酶在填充床反应器中进行循环反应 ,2 0 h左右转化率超过 40 % ,反应 3批后酶活仍保持在
Candida cylindracea (CCL) adsorbed on the macroporous resin, as well as the effect of water content and alcohol concentration on the immobilized enzyme activity has been studied. The optimal water content of immobilized CCL ranges in 12 %~23 % and the optimal octanol concentration is 40 mmol/L to naproxen 20 mmol/L. Used as immobilizing support, the weak hydrophilic macroporous resin HZ 841 can make a good micro circumstance around the lipase molecules and maintain the water content in the immobilized lipase. Three batches of circular reaction were performed on packed bed reactor (PBR) catalyzed by CCL immobilized on the weak polarity macroporous rein HZ 841. Resolution of naproxen could be achieved with conversion more than 40 %, and enzyme activity remained 98 %.
