摘要:以红霉素生产菌红色链霉菌HB为研究对象,根据红霉素生物合成机理,以促进甲基化转化和强化基础代谢为主要手段,在利用摇瓶和50L罐发酵生产红霉素的过程中,在大约127h的时候,向发酵液中流加ATP、L-Met、MgSO4和柠檬酸,采用高效液相色谱仪(HPLC)对最终发酵液进行检测,红霉素的有效组分A的相对百分含量由原来的73%提高到88%以上,对最终发酵液进行生物测定,其生物效价亦得到明显提高,实现了红霉素发酵生产的优化。
To optimize erythromycin production and it′s major components. Saccharopolyspora erythraea HB was used as producing strain. ATP, L-methionine, MgSO_4and citric acid were added to the broth at 127-hour of fermentation during the production of erythromycin in flasks and 50L fermentor. The final broth was determined by HPLC. The results showed that relative percentage of erythromycin A had been ~increased initially from 73% to more than 88%. Biological potency increased significantly. The process of erythromycin production has been optimized.
