摘要:NOVELTY - A fluorescent probe comprises a ligand portion, optionally a connector portion and a fluorescent dye portion. The fluorescent dye moiety is viscosity-responsive fluorescent dye groups including an electron donor portion, a conjugation portion and an electron acceptor moiety. The ligand moiety is capable of specifically recognizing and labeling with a target protein of a protein marker or a fusion protein. The ligand moiety A is a tomb mass that is capable of specifically recognizing and covalently labeling a target protein tagged with a protein tag or a fusion protein. USE - Fluorescent probe is used for fluorescence-activated protein-specific labeling, protein quantification, detection or kinetic studies, in cells and tissues (all claimed). ADVANTAGE - The fluorescent probe enables fluorescence-activated protein-specific labeling with high sensitivity. DETAILED DESCRIPTION - A fluorescent probe comprises a ligand portion, optionally a connector portion and a fluorescent dye portion. The fluorescent dye moiety is viscosity-responsive fluorescent dye groups including an electron donor portion, a conjugation portion and an electron acceptor moiety. The ligand moiety is capable of specifically recognizing and labeling with a target protein of a protein marker or a fusion protein. The ligand moiety A is a tomb mass that is capable of specifically recognizing and covalently labeling a target protein tagged with a protein tag or a fusion protein. The ligand moiety is directly covalently attached to the electron donor moiety of the fluorescent dye moiety, or the electron arch portion is covalently attached to the fluorescent dye moiety through the linker moiety. INDEPENDENT CLAIMS are included for the following: (1) preparation of fluorescent probe, which involves reacting an unsaturated group-containing compound of formula (II) and a ligand and optionally with a linker; (2) fluorescence-activated protein-specific labeling method, which involves contacting the fluorescent probe with a target protein, in a medium chosen from intracellular medium, organelle media, living tissue media, blood or body fluid; and (3) probe kit, which comprises the fluorescent probe, and a biocompatible medium chosen from dimethyl sulfoxide, buffer and physiological saline.
