摘要:本文对15L实验室规模发酵罐阿维菌素发酵过程主要特点,尤其是发酵液流体特性进行了研究。结果表明,阿维菌素发酵液总体为非牛顿流体,并满足剪切稀化特性,且随着发酵的进行,表观粘度和稠度系数K按一定的规律变化;而在搅拌转速一定的情况下,维持发酵液溶氧水平所需的通气量与发酵液的流变特性有紧密的关联性。较高的溶氧水平有利于获得较高效价的阿维菌素;但过高的溶氧会导致生物量迅速增长,如果控制不当,则将导致发酵液流变特性迅速恶化,并最终可能引起菌丝发糊现象的出现。因此对于高溶氧条件下的阿维菌素发酵过程,前期发酵液流体特性及反应器流场的监控非常必要。
In this paper,both the main features of avermectin fermentation process in 15L laboratory-scale fermenter and the fluid characteristics of fermentation broth were studied.The results showed that avermectin fermentation broth was non-Newtonian fluid in generally and met the shear-thinning properties.Besides,it also showed that the apparent viscosity and consistency coefficient K changed according to a certain law as the fermentation progressed.Whereas,in certain fixed stirring speed,the rheological properties of fermentation broth in the reactor were closely related with the dissolved oxygen level which was maintained by the ventilation.The high dissolved oxygen level was helpful to acquire the final titer of avermectin fermentation.However,the high oxygen can lead to the rapid growth of biomass,so the improper operation would result in rapid deterioration of the rheological properties of fermentation broth and eventually may lead to the emergence of the phenomenon of hyphal hair paste.Therefore,in high dissolved oxygen condition,the pre-fermentation liquid fluid characteristics and flow monitoring of the reactor are necessary to avermectin fermentation process control.
