摘要:研究了控制不同氧化还原电位(oxidation-reduction potential,ORP)对乳酸发酵过程的影响。通过5 L发酵罐分批发酵实验发现ORP水平控制在-170 mV时最有利于乳酸生成,乳酸最高质量浓度达176 g/L,糖酸转化率为94%,其平均乳酸产率3.7 g/(L.h),比ORP控制在-220 mV和-120 mV时分别高19%,37%;通过对发酵过程胞外有机酸浓度及代谢流分析发现氧化还原电位是通过影响细胞内代谢流分布来影响乳酸合成的。
A new method was presented by using extracellular oxidation-reduction potential (ORP) to control lactic acid biosynthesis processes by Lactobacillus paracasei. In batch fermentation, cell growth and lactic acid production were studied under three different ORPs, i.e. - 120 mV, - 170 mV, and -220 mV. The result showed that the ORP level of - 170 mV was preferable to the formation of lactic acid. At the ORP level of - 170 mV, the concentration of lactic acid reached 176 g/L and the yield based on glucose reached 94%. In addition, the productivity of lactic acid of 3.7 g/(L·h) was obtained, and it was higher than that at ORP levels of - 120 mV and -220 mV by 37% and 19%, respectively. The extracellular concentration of the organic acid was also detected and analyzed . The metabolic flux, influencing by the productivity of lactic acid was obviously affected by extracellular ORP level.
