摘要:通过小角X射线散射(SAXS)技术研究了牛血清蛋白(BSA)在阳离子球形聚电解质刷(SPB)中的吸附及分布规律。随着BSA质量浓度的增加,其在SPB中的分布均匀递增,直至达到饱和吸附;随着pH的增大,BSA在刷子层中的分布呈现从靠近核逐渐外移扩散到溶液中的变化趋势。此外,用时间分辨SAXS技术观察到在极短的时间(12s)内BSA在SPB中的动态吸附及再分布的显著变化过程。研究结果表明:SAXS技术在原位表征蛋白质在SPB中的动态分布方面具有其他表征技术不可替代的优势。
Small angle X-ray scattering (SAXS) was employed to observe the adsorption and distribution of bovine serum albumin (BSA) onto cationic spherical polyelectrolyte brush (SPB). The fitting results of SAXS data showed that the distribution of protein within brush layer increased uniformly until saturation upon increasing the BSA concentration. As pH increased, the distribution of BSA in SPB moved from the area near PS core outwards toward the solution. Moreover, time-resolved SAXS was used to observe the dynamic adsorption and re-dispersion of BSA onto cationic SPB in a very short time (12 s). Experimental results confirm that SAXS is a unique tool to observe in situ the dynamic distribution of proteins in SPB.
