摘要:[目的]测定药后茭白中敌磺钠的残留量、敌磺钠及其代谢物对2种人体细胞HepG2和HEK-293的毒性,为该药物在茭白上的使用安全性提供技术资料。[方法]茭白在药后1、3、5 d采样,用QuEChERS方法前处理,GC-MS方法测定其代谢产物N,N-二甲基苯胺在茭白食用部分的残留。利用MTT法测定敌磺钠及其代谢产物N,N-二甲基苯胺对2种人体细胞HepG2和HEK-293的24 h毒性作用。[结果]喷药后第1天N,N-二甲基苯胺的残留检测量为147.08μg/kg,第3天和第5天并未检测到残留;敌磺钠对HepG2和HEK-293的IC50值分别是166.77、309.76 mg/L,N, N-二甲基苯胺对2种细胞的IC50值分别是788.19、956.72 mg/L。[结论]敌磺钠及其代谢物在茭白内残留量低,对细胞HepG2和HEK-293毒性低,为敌磺钠安全性研究提供了科学依据。
[Aims] This study aims to detect the residue of fenaminosulf and its metabolite N,N-dimethylaniline in Zizania latifolia and the cytotoxicity to two kind of human cells HepG2 and HEK-293, to provide technical information on the safety of the pesticide in the use of Z. latifolia. [Methods] Z. latifolia samples were taken on the first, third and fifth days after treatment, by QuEChERS method to process samples and GC-MS to detect the residue of its metabolites N,N-dimethylaniline in Z. latifolia. The MTT assay was used to determine the 24 h cytotoxicity on the cell HepG2 and HEK-293. [Results] The residue of N,N-dimethylaniline on the first day after treatment was 147.08 μg/kg, no residue was detected on the third day and the fifth day;the IC50 of HepG2 and HEK-293 treated with fenaminosulf are166.77, 309.76 mg/L, the IC50 of HepG2 and HEK-293 treated with N,N-dimethylaniline are 788.19, 956.72 mg/L.[Conclusions] The residue of fenaminosulf and its metabolite N,N-dimethylaniline in Z. latifolia is low, and the cytotoxicity to cells HepG2 and HEK-293 is low. It provides scientific basis for study on safety of fenaminosulf.
