摘要:通过动态吸附和解吸附研究,从5种色谱填料中筛选出适合唾液酸乳糖与乳糖分离的色谱树脂,建立了唾液酸乳糖和乳糖的色谱分离技术,考察了径高比、洗脱速度、柱温、上样浓度等色谱条件对分离度和回收率的影响,确定了最佳色谱分离条件.结果表明,树脂QY-Z001可高效分离唾液酸乳糖和乳糖,最佳分离条件为径高比1:35、温度55℃、流速1.0 m L/min.、进样浓度100 g/L.该条件下唾液酸乳糖与乳糖分离度可达0.96,唾液酸乳糖含量由6.8%提高至84.6%,回收率为76.9%.
A resin was selected from five kinds of Chromatographic packings through dynamic adsorption process and desorption process to establish a chromatographic separation method to further purify sialyllactose from the crude extract,which was suitable to separation sialyllactose and lactose.Then the effects of ratio of diameter to length,temperature,flow rate and the concentration were studied on the recovery rate and resolution,finally,the optimum conditions for separation of sialyllactose were determined.The results showed that the resin QY-Z001 can efficiently separate sialyllactose and lactose,under the optimum conditions:ratio of diameter to length 1:35,temperature of 55℃,1.0 mL/min flow rate,and the sample concentration 100 g/L.At the optimum conditions the resolution of sialyllactose and lactose can reach 0.96,and purity of sialyllactose increased from 6.8%to 84.6%,with a recovery rate 76.9%.
