摘要:Platelet-rich plasma (PRP) is widely used in various medical treatments; however, its clinical effect is scattered because of time-consuming separations. Here, a sodium alginate (SA)-based thixotropic gel was produced by internal gelation of calcium, yielding a "gel-slit" dependent quick (within 4 min) and gentle (135 g) PRP separation. By analyzing the rheological properties of gels with different SA concentrations and calcium cross-linking degrees, it was found that the gels' viscosity thinning rate (reaching a proper viscosity under centrifugal shear for red blood cell sedimentation) during shearling (or centrifugation) is a key parameter determining the separation efficacy and PRP quality. A thixotropic gel, made from 8 % of SA and a calcium cross-linking degree of 0.4, has an optimal viscosity thinning rate (the viscosity thins 63.99 f 0.31 % to reach a viscosity of 53.38 f 0.95 Pa & sdot;s) for quick PRP separation at low centrifugation speed. By spinning at 1100 revolutions per minute (135 g) for merely 4 min, the gel enables high yield (production of 1.32 f 0.03 mL PRP from 3 mL of whole blood) and high platelet capture efficiency (80.84 f 1.31 %) PRP separation (the platelet concentration was1.84 f 0.02 times that of the original blood). This study provides new insights into the mechanism of thixotropic gel-assisted PRP separation for more convenient PRP treatments.
