摘要:胞外几丁质酶等多种水解酶系在虫草侵染宿主的过程中发挥着重要的作用,增强酶活对于提高虫草子实体人工培养的成功率有着重要的意义。文中研究考查了多种碳、氮源、初始培养基pH值和培养时间对于液态培养的蛹虫草菌丝体胞外几丁质酶活性的影响。结果表明,几丁质酶的活性主要取决于碳源,几丁质对几丁质酶的诱导合成十分重要,其中以1%胶状几丁质最佳。当培养基中不存在几丁质、细胞优先利用其它营养物质或存在酶解产物阻遏时,酶的合成就会被抑制。最终确定的优化培养基以2%胶状几丁质为碳源,0.1%酵母膏为氮源,初始培养基pH值6.0;培养时间5d,此时胞外几丁质酶的活性为12.0mU/mL以上。
The extracellular chitinase is one of the crucial factors for Cordyceps militaris in infecting hosts; thus, increasing its activity will be helpful to the development of artificial C. militaris fruit body cultivation. In this study, the abilities of C. militaris for chitinase production under multiple conditions (carbon sources, nitrogen sources, initial pH of medium and fermentation time) were explored. Results showed that the production of chitinase mainly depended on carbon sources, and 1% colloidal chitin was the best inducer of chitinase. If the inducer was removed, or C. militaris preferentially utilize other nutrition, or the end products of chitinase reaction were excessive, the enzyme synthesis will be inhibited. The most suitable medium for accumulation of chitinase was found to be 1% colloidal chitin, 0.1% yeast extract, pH 6.0% the duration of fermentation was 5 days. Under optimized condition, the extracellular chitinase activity was increased to 12mU/mL.
