摘要:采用开环聚合及酯化反应得到了末端含有卟啉修饰的两亲性嵌段共聚物PEG(113)-PCL(46)-卟啉,并采用~1HNMR、GPC对其结构进行了表征;利用分光光度计测定其临界胶束浓度(CMC);选用透析法制备了嵌段共聚物的空白及载紫杉醇(PTX)药物胶束,并采用粒度仪及透射电镜对胶束粒径及形态分布进行了测试。此外,还考察了胶束的载药量及体外释放速率。结果表明:由于共聚物末端含有的卟啉能与PTX形成非共价键,不仅PTX的载药量有明显提高,而且体外释放时间得以延长。
The amphiphilic block copolymer,PEG(113)-PCL(46)-porphyrin was first synthesized by using ring-opening polymerization and then modified with porphyrin via esterification reaction.~1HNMR and GPC was employed here to characterization these block copolymers. The critical micelle concentration( CMC) of these block copolymers were tested by spectrophotometer. PTX free or loaded micelles were obtained by dialysis method. The size and polydispersity index( PDI) was checked with particle size analyzer. Transmission electron microscope( TEM) was also used here to obtain the morphology of the PTX free and loaded PEG(113)-PCL(46)-porphyrin micelles. Besides that,the drug loading capacity and release rate in vitro were tested. The results showed that PEG(113)-PCL(46)-porphyrin have higher PTX loading capacity and a longer drug release time than that of PEG(113)-PCL(46).
