摘要:目的研究南海海域Euplexaura rhipidalis种属柳珊瑚的丙酮提取物及其不同溶剂萃取部位的抗肿瘤活性,探讨最强活性部位可能的作用机制。方法丙酮提取物及其萃取部位作用于人肺癌细胞A549和人肝癌细胞Hep G2,采用磺酰罗丹明B(SRB)法检测细胞增殖抑制率;甲醇重溶部位(EM)以低、中、高浓度作用于A549和Hep G2细胞,采用流式细胞仪检测细胞凋亡比率和细胞周期分布。结果 EM的体外抗肿瘤活性最强,对A549和Hep G2细胞的IC50值均小于10μg·ml-1,优于或接近阳性对照5-FU;EM能显著诱导细胞凋亡并呈现良好的浓度依赖性,中高浓度时对细胞G2/M或S期有周期阻滞作用。结论甲醇重溶部位是柳珊瑚Euplexaura rhipidalis主要的抗肿瘤活性部位,能抑制A549和Hep G2细胞的增殖,具有诱导凋亡、阻滞周期的作用。
Objective To investigate the antitumor fraction of the Euplexaura rhipidalis extracted by different solvent and to explore the potential mechanism of its action. Methods The anti-prolife ration of the acetone extract and its fractions extracted by different solvent against A549 and HepG2 cell lines has been investigated by Sulforhodamine B( SRB) method. Meanwhile,the effects of the methanol re-solvent fraction( EM) on cell apoptosis and cell cycle arrest were also studied by flow cytometry method( FCM). Results The EM had the strongest anti-proliferative effect on A549 and HepG2 cells in vitro,of which IC50 values were less than 10 μg·ml-1and were close to or lower than that of the positive reference 5-FU. Additionally,EM could cause A549 and HepG2 cell apoptosisin a dose-dependent manner,and could induce G2/M or S phase arrest at medium and high concentrations. Conclusion The EM is considered as antitumor fraction of the Euplexau rarhipidalis against A549 and HepG2 cells.Its anti-proliferative activity may be attributed to the cell apoptosis and cell cycle arrest.
