摘要:[目的]建立同时检测茭白中吲哚乙酸(IAA)、玉米素(ZT)、玉米素核苷(ZR)、脱落酸(ABA)、赤霉素(GA3)等5种内源性植物激素的超高效液相色谱-串联质谱(UHPLC-MS/MS)方法。[方法]样品经BHT-甲酸-甲醇溶液提取,分散固相萃取方法(PSA)净化,采用超高效液相色谱-串联质谱法进行分析检测,基质标准曲线外标法定量分析。[结果]该方法在0.0002~0.1 mg/L范围内,具有良好的线性关系,决定系数大于0.99。在0.005、0.025、0.1 mg/kg添加水平下,5种内源植物激素的回收率为80.3%~116.0%,相对标准偏差为1.9%~9.1%。5种激素方法的定量限在0.0002~0.005 mg/kg。[结论]该方法简便、快速、灵敏、准确,为快速准确地分离和测定茭白不同部位内源激素提供了有效方法。
[Aims] This study aims to develop a ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS) method for the determination of endogenous hormones in Zizania caduciflora. [Methods] The Z.caduciflora samples were extracted with methanol-formic acid(99:1, by vol, 1 mmol/L BHT), and purified by dispersive solid phase extraction method using PSA sorbent, and then analyzed by liquid chromatography-tandem mass spectrometry(LC-MS/MS), the extraction solvent, extraction method and purification agent were optimized. [Results] The results showed that the calibration curves of five endogenous hormones were linear(R2≥0.99) in the concentration range of 0.0002-0.1 mg/L. The average recoveries at three spiked levels of 0.005, 0.025 and 0.1 mg/kg were in the ranges of 80.3-116.0%, with relative standard deviations(RSDs) of 1.9-9.1%. The limits of quantification(LOQ) for all analytes were between 0.0002 and 0.005 mg/kg. [Conclusions] The results showed that the method was simple, rapid,sensitive and accurate. It provided an effective method for rapid and accurate separation and determination of endogenous hormones in different parts of Z. caduciflora.
