摘要:本试验以巯基乙酸(TGA)作为稳定剂合成水溶性CdTe量子点(QDs),采用斑马鱼胚胎发育技术,观测0、1、25、50、100、200、300、400nmol·L-1不同浓度的水溶性CdTe量子点对斑马鱼胚胎发育的影响.记录胚胎发育过程中具有代表性的毒理学终点(自主运动频率、内心率、体长).结果表明:在斑马鱼120hpf(hours post fertilization)胚胎受精后,TGA-CdTe量子点半数致死浓度(LC50)为185.9nmol·L-1.量子点TGA-CdTe对斑马鱼胚胎24hpf时1min自主运动频率、48hpf时10s内心率、孵化率、体长都存在剂量-效应关系.TGA-CdTe能引起斑马鱼胚胎发生多种中毒症状,如心包囊肿、卵黄囊肿、脊柱弯曲、体节减少、心率减慢、黑色素沉积少等异常现象.
The effects of various concentrations (0、1、25、50、100、200、300、400 nmol·L-1) of water-soluble CdTe quantum dots (QDs) synthesized using mercaptoacetic acid (TGA) as a stabilizer on the embryonic development of zebrafish (Danio rerio) were investigated in this study.The typical morphological,physiological,and behavioral endpoints were recorded in the embryonic development period.The results showed that the median lethal concentration (LC50) for zebrafish embryos exposed to TGA-CdTe QDs after 120 hpf (hours post fertilization) was 185.9 nmol·L-1.The TGA-CdTe QDs dosage-response relationships existed between the frequency of spontaneous movement in 1 min after 24 hpf,the frequency of heart beat in 10 s after 48 hpf,the hatching rate and the body length.Several abnormalities and toxic symptoms caused by TGA-CdTe QDs could be observed including pericardial cyst,vitelline cyst,spinal curvature,defects of somite development,decreased heart rate and less melanin pigmentation.
