摘要:用高效液相色谱法测定魔芋中葡甘露聚糖 (KGM)的含量。色谱柱为 Spherisorb-NH2柱 ,柱温 40°C,流动相为乙腈 -Na H2 PO4 (0 .0 1 2 mol/ L) (4∶ 1 ,V/ V) ,示差折光检测器检测 KGM水解液中的甘露糖 (M)和葡萄糖 (G)。KGM参比样品的回收率为 96.3 %。用 M、G的标准品测定魔芋精粉中 KGM的含量为 5 8.5 % ,KGM分子中甘露糖和葡萄糖的摩尔比值为 1 .60。本方法可用于魔芋生产以及其他产品中的
Konjac glucomannan was determined by HPLC. Mannose and glucose in hydrolytic solution were seperated on a Spherisorb NH 2 column, with column temperature 40 °C and mobile phase of acetonitrile sodium dihydrogen phosphate (0.012mol/L)(4∶1, V/V ), and were detected by differential refractometer. The average recovery of KGM is 96.3%. The content of KGM and the ratio of mannose to glucose in KGM were calculated. This method has been applied to analyze hydrolytic solutions of KGM from different sources.
