摘要:建立了液相色谱串联质谱法同时测定鸡蛋中甲硝唑及新代谢物的方法。通过液相色谱串联高分辨质谱进行定性扫描,在鸡蛋中发现一种新的甲硝唑代谢物-羧基甲硝唑。进一步建立了以0.1%(V/V)甲酸乙腈溶液提取,石油醚净化,Waters Atlantis T3(100 mm×2.1 mm, 3μm)柱分离,正离子扫描和多反应监测模式采集,液相色谱串联质谱法测定鸡蛋中甲硝唑及其代谢物的定量方法。结果表明,在质量浓度0.50~20.0 ng/mL范围内,甲硝唑及其代谢物呈良好的线性关系,检出限为0.25μg/kg,定量限为0.5μg/kg;平均加标回收率在85.5%~110.9%之间,相对标准偏差在0.9%~13%之间。该方法能够应用于实际鸡蛋样品中甲硝唑及代谢物的检测。
A method for the simultaneous determination of metronidazole and new metabolites in eggs by liquid chromatographytandem mass spectrometry was established in this study.A new metronidazole metabolite,carboxyl metronidazole,was found by qualitative scanning of liquid chromatography tandem high-resolution mass spectrometry.Further,a quantitative method was established with liquid chromatography tandem mass spectrometry for the determination of metronidazole and its metabolites in eggs by 0.1%(V/V)formic acid acetonitrile extraction,petroleum ether purification,Waters Atlantis T3 column(100 mm×2.1 mm,3μm)separation,positive ion scanning and multi-reaction monitoring mode collection.The results showed that metronidazole and its metabolites showed good linear relationships in the range of 0.5020.0 ng/mL.The limits of detection and quantification were 0.25μg/kg and 0.5μg/kg,respectively.The average recoveries were 85.5%110.9%,and the relative standard deviations were 0.9%13%.Additionally,this method can be applied to the detection of metronidazole and metabolites in actual eggs samples.
