摘要:采用HADAMARD统计设计方法建立了适于重组BHK细胞培养和vWF蛋白表达的无血清培养基SFMA。用SFMA进行重组BHK细胞的批培养,细胞生长明显优于有血清培养(DMEM+FBS(φFBS=0.05))和无血清培养基SFM- 的培养,最大细胞密度可分别提高16.7%和40%。同时,在培养的96h,蛋白的表达水平比有血清培养提高了18.7%,SFMA维持培养48h的vWF蛋白的表达水平比SFM- 提高了50%。
A new serum-free medium SFMA was formulated using HADAMARD statistical design for the cultivation of rBHK cells and the expression of rec-vWF. Comparing with that in (DMEM+FBS(φ_(FBS)=0.05)) and SFM-Ⅱ, rBHK cells grew better in SFMA, and the maximum cell density increased by (16.7%) and 40%, respectively. Moreover, the expression of rec-vWF in SFMA was 18.7% higher than that in (DMEM+FBS(φ_(FBS)=0.05)), and the concentration of rec-vWF in SFMA was also 50% higher than that in SFM-Ⅱ in maintaining cultures of rBHK cells in serum-free media.
