摘要:目的:优化壳聚糖絮凝法制备双孢蘑菇多糖(Agaricus bisporus polysaccharides,ABP)工艺并研究其抗氧化活性。方法:以多糖回收率、蛋白质脱除率和脱色率为指标,采用层次分析法(Analytic hierarchy process,AHP)建立综合评分体系,在正交试验的基础上优化絮凝工艺。以水提醇沉多糖(Ethanol precipitated-ABP,E-ABP)作为对照,评价壳聚糖絮凝多糖(Chitosan flocculated-ABP,CF-ABP)的抗氧化活性。结果:最佳工艺条件为壳聚糖添加量2%、温度45℃、时间120 min、pH值为4,CF-ABP得率为(7.64±0.49)%,纯度为(60.76±0.70)%。CF-ABP和E-ABP的1,1-二苯基-2-三硝基苯肼(1,1-Diphenyl-2-picrylhydrazyl,DPPH)自由基清除能力的IC_(50)值分别为1.468、7.246 mg/mL,两者羟自由基清除能力的IC50值分别为0.122、0.216 mg/mL,在4 mg/mL浓度下CF-ABP和E-ABP的总还原力分别为0.51和0.27。结论:壳聚糖絮凝法可用于制备双孢蘑菇多糖,得到的CF-ABP抗氧化活性优于E-ABP。此结果可为双孢蘑菇多糖的制备提供新的工艺参考。
Objective:To optimize the preparation process of polysaccharides from Agaricus bisporus by chitosan-based flocculation and investigate its antioxidant activity.Methods:A comprehensive scoring system was established with analytic hierarchy process(AHP)with polysaccharide recovery rate,protein removal rate and decolorization rate as the indicators,and the orthogonal method was used to optimize the flocculation conditions.The antioxidant activity of ethanol precipitated polysaccharide(E-ABP)and chitosan flocculated polysaccharide(CF-ABP)was measured and E-ABP was taken as the control.Results:The optimum flocculation conditions were as follows:2%flocculant addition,45℃,120 min,pH 4.The yield and purity of CF-ABP was(7.64±0.49)%and(60.76±0.70)%,respectively.The IC50 values of the 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical scavenging capacity of CF-ABP and E-ABP were 1.468 mg/mL and 7.246 mg/mL,respectively.And the IC_(50)values of the hydroxyl radical scavenging capacity of CF-ABP and E-ABP were 0.122 mg/mL and 0.216 mg/mL,respectively.With a concentration of 4 mg/mL,the total reducing power of CF-ABP and E-ABP were 0.51 and 0.27,respectively.Conclusion:Chitosan flocculation method can be used for the preparation of polysaccharide from Agaricus bisporus,and the CF-ABP shows better antioxidant activity compared to E-ABP.This result provides a new reference for the process of preparation of polysaccharide from Agaricus bisporus.
