摘要：目的：探讨灵芝孢子油对人肺癌细胞（A549）超微结构的影响以及对其生长的抑制作用.方法：利用CCK-8法检测灵芝孢子油对A549细胞的生长抑制作用;利用透射电镜研究灵芝孢子油对A549细胞超微结构的影响;利用乳酸脱氢酶（LDH）检测法研究灵芝孢子油对细胞膜通透性的影响;上述方法研究灵芝孢子油对体外培养的肺癌细胞A549的抑制作用.结果：CCK-8检测结果表明25 μg/ml浓度的灵芝孢子油可导致A549细胞的存活率明显下降;透射电镜结果显示,随着灵芝孢子油浓度的增加,可以观察到细胞超微结构的明显改变;LDH检测显示灵芝孢子油可使细胞的LDH释放量明显增加,且呈剂量依赖性.结论：灵芝孢子油对人肺癌细胞具有明显抑制作用.
Objective:To study the effect of ganoderma lucidum spore oil on growth inhibition and ultrastructure of human lung cancer cells(A549).Methods:The effect of growth inhibition was detected by cell counting kit 8(CCK-8) method.The ultrastructure of A549 cells was observed by transmission electron microscopy.The viability of A549 cells was studied by CCK-8 test.The membrane permeability of A549 cells was studied by lactate dehydrogenase(LDH) cytotoxicity assay.Results:Ganoderma lucidum spore oil significantly decreased the viability of A549 by CCK-8 in 25μg/ml.The ultrastructure of A549 was changed and LDH release of A549 had a significant increase with a concentration-dependent relationship.Conclusion:Ganoderma lucidum spore oil can inhibit the viability of human lung cancer cells.
