摘要:研制了适合纯化猪血超氧物岐化酶(Cu.Zn-SOD)溶液的醋酸纤维素(CA)超滤膜,其截留分子量约为16 000。研究了溶液浓度、操作压力等因素对通量的影响。结果表明,自制CA膜超滤纯化SOD溶液性能优于一些市售膜。收率为86~89%,比活提高25%,纯化因子1.35。利用透析过滤法,基本上能除去溶液中游离铜离子。
A cellulose acetate ultrafiltration membrane with molecular weight cut-off of 16000 was prepared and tested for the purification of pig blood superoxide dismutase (Cu. Zn-SOD) from its solution. The effects of the concentration and the operation pressure on flux were investigated. With a yield of 86~89%, a raise of specific activity of SOD by 25% and a purification rate of 1.35, the results showed that the CA ultrafiltration membrane was better than commerical ones. The free cupric ions in the solution were basically removed by diafiltration,
