摘要:建立了免疫磁珠-超高效液相色谱串联质谱检测黄曲霉毒素B_(1)(AFB_(1))的方法。分子对接研究表明黄曲霉毒素B_(1)(AFB_(1))与大豆蛋白间存在较强的相互作用;实际样本分析显示食品基质中存在的干扰物严重影响测定结果的准确性。本研究以琼脂糖Protein A/G磁珠固定抗AFB_(1)抗体,制备了AFB_(1)免疫磁珠。免疫磁珠的抗体固定量为7.95μg/μL;每20μL免疫磁珠对AFB_(1)的最大吸附量为0.548μg。以10%乙酸乙腈溶液作为洗脱液,以超高压效液相色谱串联质谱(UHPLC-MS/MS)法对AFB_(1)进行测定。在0.06~10μg/mL范围内,质谱信号与AFB_(1)浓度呈线性关系,检出限为0.033μg/mL。采用该免疫磁珠对玉米淀粉进行净化前处理,玉米淀粉中AFB_(1)的加标回收率为94.0%~105.9%。该免疫磁珠可用于食品淀粉抽提液中AFB_(1)的富集净化。
A method using immunomagnetic beads coupled with ultra-high performance liquid chromatographytandem mass spectrometry was developed for the determination of aflatoxin B.Molecular docking study shows that there is strong interaction between aflatoxin B_(1)(AFB_(1))and soy proteins.However,actual sample analysis shows that the presence of interfering substances in food matrices affects the accuracy of the results.Agarose protein A/G magnetic beads were selected to immobilize anti AFB_(1) antibody to prepare AFB_(1) immune magnetic beads.The antibody immobilization capacity of immunomagnetic beads is 7.95μg/μL,and the maximum adsorption capacity of AFB_(1) per 20μL of immunomagnetic beads is 0.548μg.After eluted with 10%acetonitrile acetate,AFB_(1) could be determined by ultra-high performance liquid chromatography-tandem mass spectrometry(UHPLC-MS/MS).The mass spectrometric signal was linear with AFB_(1) concentration in the range of 0.06-10μg/mL,with the detection limit of 0.033μg/mL.The spiked recoveries of AFB_(1) in corn starch purified by this immunomagnetic beads were 94.0%105.9%.The immune magnetic beads can be applied to the purification and enrichment of AFB_(1) in food starch extracts.
