摘要:以文献报道的新型荧光探针MNP为衍生化试剂,对半胱氨酸(cysteine,Cys)、同型半胱氨酸(homocysteine,Hcy)和谷胱甘肽(glutathione,GSH)进行柱前衍生化处理,并利用HPLC-UV法对三种硫醇衍生物进行分离分析。实验结果显示MNP与GSH/Cys衍生化产物的保留时间分别为5.9min和7.1min,表明采用衍生化法可实现对GSH和Cys的分离。虽然Cys和Hcy的结构非常相似,通过优化分离条件可以实现对两者的部分分离。定量结果表明GSH/Cys衍生化产物的峰面积与在1~30μM/1~20μM范围内与硫醇的浓度呈线性关系,该法对硫醇的检测限约为20nM。
Cysteine (Cys), homocysteine (Hcy) and glutathione (GSH) were separated and analyzed by HPLC-UV using pre-column derivatization with the reported probe MNP as the derivatization reagent. The results showed that Cys and GSH could be separated well with the retention times of their derivatized products being 5.9 min and 7.1 min, respectively. However, only partial separation were obtained between the derivatives of Cys and Hcy because of their similar structures. Good linear relationship between the peak area and GSH/Cys concentration were found in the range of 1~30 μM/ 1~20 μM. The detection limit of GSH/Cys was calculated to be about 20 nM.
