摘要:目的 确定重组人锰超氧化物歧化酶 (rhMn SOD)工程菌高效表达的最适培养工艺条件。方法 通过摇瓶培养研究了包括接种量、诱导时机、Mn2 + 浓度、诱导后培养时间、发酵过程中 pH变化以及初始 pH值对发酵的影响。 结果 该工程菌的最佳接种量为 3%,最佳诱导时机为接种后 3h ,确定Mn2 + 浓度为 6 0 0 0 μmol·L-1,诱导后培养 5h ,在培养过程中当pH为6 .83时外源蛋白可获得高效表达 ,初始 pH确定为 8.0。 结论 :诱导和培养基的 pH是影响工程菌发酵的主要因素 ,尤其是Mn2 + 的加入表达有活性的rhMn SOD是必需的。
OBJECTIVE: To define the suitable culture technology of recombinant human maganese-superoxide dismutase. METHODS: The conditions including inoculum quantities, induced time, concentration of Mn2+, culture time after induced, changes of pH during the culture and original pH of culture by culture in shaking-bottle were investigated. RESULTS: The suitable culture technology conditions were 3 % inoculum quantities, induced by 6 000 μmol·L-1 Mn2+ for 3 hours after inoculum, and cultured 5 hours after induced. The exogenous protein expressed highly at pH 6.83 and the original pH was 8.0. CONCLUSION: Induction and pH is the key factor that affect the fermentation of the engineer bacterium, especially the additition of the Mn2+ is necessary to the expression of the active rhMn-SOD.
