摘要:本文研究了添加甘油对北里链霉菌ZY150504发酵吉他霉素效价和A5组分的影响。考察了甘油的添加浓度和添加时间对发酵液效价的影响,发现通过添加甘油,提高发酵培养基中的起始甘油浓度,有效地提高了吉他霉素的效价。发酵结束时发酵液的效价为10847U/m L,比对照提高了47%,比文献报道效价提高了27%;吉他霉素A5组分含量提高17%;而A4组分含量降低了36%。进一步研究发酵过程发现添加甘油之后,发酵过程中条件组的脂肪酶活力较高,油脂利用较快,丙酸含量增加,乙酸浓度较低,促进了吉他霉素的生物合成,提高了A5组分的含量。
Effects of the addition of glycerol on the kitasamycin titers and components of Streptomyces kitasatoensis ZY150504 fermentation were investigated. The concentration and adding time of glycerol was changed to study the best adding condition. The results showed that adding glycerol which increased the glycerol concentration of the beginning of the fermentation medium. The final titer of kitasamycin reached to 10847U/mL, which was 47% higher than the control and 27% higher than the report, the content ofA5 component increased 17% and the content of A4 component decreased 36% comparing to the control. The lipase activity was higher by adding glycerol, making soybean oil decomposed quickly. The higher content of propionic acid and the lower concentration of acetic acid promoted the biosynthesis of kitasamycin and increased the concentration ofA5 component.
