摘要:研究了采用大肠杆菌游离细胞核苷磷酸化酶合成核苷药物过程中反应平衡常数和底物转化率的计算方法及其变化规律,阐明了游离细胞内核苷磷酸化酶的活力与细胞的培养、菌体的预处理方式密切相关。实验结果表明,大肠杆菌湿菌体经4°C干燥处理2~3d后,胞内核苷磷酸化酶不易失活,并能有效地降低胞内抑制物对酶反应的抑制作用。与湿菌体相比,干燥菌体用于酶反应时底物的转化率可提高33.5%。
The calculation method of theoretical substrate conversion rate in the enzyma tic preparation of nucleoside pharmaceuticals catalyzed by phosphorylase from free E.coli cells was studied.In addition,It was shown that phosphorylase activity of free cells was closely related to their culture condition and pretreatment procedures.If wet E.coli cells were dried at 4°C 2~3 days,the stability of phosphorylase within cells was increased,the inhibition within cells reduced and the substrate conversion rate increased by 33.5%.
