摘要:目的:优化TATm-survivin(T34A)蛋白的冻干工艺,同时对该蛋白的体外抗肿瘤活性进行研究。方法:采用MTT法检测TATm-survivin(T34A)蛋白的抗肿瘤活性并以此为指标优化该蛋白的冻干工艺。结果:海藻糖、甘露醇等的浓度对于TATm-survivin(T34A)蛋白冻干产品的外观影响很大,不同的预冻方式对冻干产品的活性有很大的影响;TATm-survivin(T34A)蛋白对于肺癌A549细胞以及乳腺癌MCF-7/s细胞有着显著的抑制效果,其中肺癌A549的效果要强于用于对照的乳腺癌B-Cap-37细胞以及胰腺癌SW1990细胞;0.25个单位的TATm-survivin(T34A)蛋白与阿霉素联合用药能够显著提高阿霉素对B-Cap-37,MCF-7/s和耐药株MCF-7/ADR的抑制效果,联合用药对这3种细胞株的IC50值分别可以降低26.6%,35.5%和18.0%。结论:获得了一条经过优化的TATm-survivin(T34A)蛋白冻干产品的制备工艺。并证明TATm-sur-vivin(T34A)蛋白具有较强的抗肿瘤效果,同时还发现该蛋白对阿霉素具有较强的增敏作用,是一种值得进一步研究与开发的抗肿瘤药物。
Objective: To develop a freeze-drying process of recombinant protein TATm-survivin (T34A), and to study its in vitro anti-tumor activities. Methods: MTT reduction assay was used to detect anti-tumor effect and as an indicator for optimizing freeze-drying process. Results: Trehalose and mannitol had great influences on the appearance of TATm-survivin (T34A) lyophilized product. TATm-survivin (T34A) had a remarkable anti- tumor effect on A549 and MCF-7/s cells, especially on A549 cells. Furthermore, when combined with adriamycin ( ADM), 0.25 U of TATm-survivin (T34A) restored a strong anti-tumor effect of ADM. The reduction rates of ICs0 values were 26.6% , 35.5% and 18.0% in B-Cap-37, MCF-7/s and MCF-7/ADRcells. Conclusion: TATm-survivin (T34A) prepared by the optimized freeze-drying process has more potent anti-tumor activities in vitro.
