摘要:本文采用大肠杆菌核苷磷酸化酶,以5-氟尿嘧啶(FU)、2'-脱氧尿苷(dU)和磷酸盐为底物酶法合成2'-脱氧-5-氟尿苷,实验结果表明,含10mmol/LdU、30mmol/LFU、10-30mmol/L磷酸盐的混合物加入5%培养24h的大肠杆菌湿菌体于pH7.0反应3h,底物dU的转化率可达45.9%。
he enzymatic synthesis of2'-deoxy-5-fluoro-urluine with E. coli nucleoside phosphorylase was stud-ied. The substrates of it were 5-fluorouracil,2'-deoxy-uridine and potassium phosphate. The experi-ment showed that the optimum substrates concentration was 10 mmol/L dU , 30 mmol/L FU,10-30mmol/L potassium phosphate. When we added 5%(w/V) E. coli wet cell cultured for 24h to the re-acting mixture and adjusted pH to 7.0 with KOH, the biosynthesis took place at 70℃ for 3 h effective-ly and 45.9%dU was converted to FdU.
