摘要:建立了测定发酵液中红霉素组分的高效液相色谱法。借鉴常用的红霉素成品高效液相分析方法,对流动相的缓冲液系统进行了优化,发现以(0.025mol/LK2HPO4)∶(乙腈)=60∶40作为流动相,红霉素A、B、C组分能够达到很好的分离效果。改进了红霉素发酵液的预处理方式,采用冷冻干燥法处理发酵液样品,能降低预处理的样品损失率,更有效地去除杂质,在保证准确度和分离效果的同时,减少了发酵液样品量,并可降低有机溶剂使用量。
A HPLC method for the determination of erythromycin components in fermentation broth was established. The constituents of the mobile phase were optimized based on the HPLC method used for analyzing pure erythromycin. Erythromycin A, B and C were separated well by reversed-phase column with 0. 025 mol/L potassium phosphate buffer and acetonitrile (60: 40) as mobile phase. In addition, lyophilization was applied to prepare the sample from fermentation broth for HPLC. This novel pretreatment method reduced the loss percent of samples during the process and removed the impurity in the fermentation broth. Thus, erthromycins could be separated effectively and deected accurately with the less consumption of organic solvent and the less amount of fermentantion broth required.
