摘要:提取棘孢小单孢菌绛红变种的细胞壁和肽聚糖。肽聚糖是细胞壁的主要成分 ,吸附的庆大霉素占细胞壁吸附总量的 95 %左右。肽聚糖吸附庆大霉素的最适 pH是 4~ 9,当溶液中没有自由状态的庆大霉素存在时 ,肽聚糖最多能吸附庆大霉素约为 880 0 0u/ g肽聚糖 (干重 )。金属离子和胺类化合物都能够竞争肽聚糖上的庆大霉素吸附位点 ,镁离子和钠离子在庆大霉素浓度分别小于 10 0 0u/ml和大于 2 0 0 0u/ml时解吸附效果最好 ,Tris也具有一定的解吸附作用。
Cell wall (CW) and peptidoglycan (PG) were isolated from Micromonospora echinospora. PG, the major part of cell wall, can bind about 95% of the total GM adsorbed on the cell wall. The optimum pH appeared to be 4~9 for the binding of GM by peptidoglycan. The maximum content of bound GM was ca. 88000u/g of dry weight of peptidoglycan and no any free GM was detected in the solution. Metal ions and amine compete with GM for the binding site. Mg 2+ and Na+ desorbed the binding of GM intensively at GM concentration less than 1000u/ml and more than 2000u/ml, respectively. Tris can also release GM from peptidoglycan to a certain degree.
