摘要:以得率为指标,通过单因素分组试验考察了液固比、粒径大小、微波辅助时间、提取级数等因素对猴头菇总多糖提取工艺的影响;通过正交试验确定了絮凝纯化的工艺参数;测定了最佳工艺条件下的猴头菇总多糖溶液对羟基自由基的清除能力.研究结果表明:最佳的提取分离工艺条件为液固比25mL/g,粒径20~40目,微波辅助时间210s,提取级数Ⅰ级,絮凝温度60℃,絮凝剂壳聚糖用量1.0mL/g,絮凝作用时间2.5h.采用微波提取得到的总多糖对羟基自由基的清除能力优于水提醇沉法.
Using extraction rate as evaluation index,the extraction parameters including liquid to solid ratio,particle size,microwave-assisted time and stages,were studied through the single factor test systematically.The flocculation purification process parameters were obtained by orthogonal experiments.The total polysaccharides solution from Hericium erinaceus under optimum conditions was also studied for hydroxyl radical scavenging activity.The optimum condition of extraction and purification process was obtained as follows:liquid to solid ratio was 25mL/g,particle size was 20~40mesh,microwave-assisted time was 210 s,extraction progression was 1stage,flocculation temperature was 60 ℃,flocculant chitosan dosage was 1.0mL/g,flocculation time was 2.5h.So the scavenging activity in vitro of polysaccharides obtained by microwave extraction-flocculation purification process was higher than that of hot water reflux extraction-alcohol precipitation.
