摘要:建立了亲水相互作用色谱法(HILIC)同时测定中度嗜盐菌中2-甲基-1,4,5,6-四氢嘧啶-4-羧酸(简称四氢嘧啶)和其羟基化衍生物(简称羟基四氢嘧啶)的方法。以乙腈和NH4H2PO4缓冲溶液为流动相,在210 nm的检测波长下,用ZIC-HILIC柱进行分离,可同时测定提取样品中的四氢嘧啶和羟基四氢嘧啶。结果表明,四氢嘧啶和羟基四氢嘧啶的质量浓度与峰面积在4.7-470 mg/L范围内呈现良好的线性关系,相关系数均为0.9999。其日内精密度分别为0.36%和0.42%,日间精密度分别为1.0%和2.4%,检出限分别为0.061 mg/L和0.065 mg/L,定量限分别为0.23 mg/L和0.25 mg/L,平均加标回收率在89.6%-100.7%之间。
A hydrophilic interaction liquid chromatography method and hydroxyectoine in moderately halophilic bacteria. The extracted ZIC-HILIC column successfully by using acetonitrile and NHaH2PO4 detector under the wavelength of 210 nm. For ectoine and hydroxyec was developed for determination of ectoine samples were separated on Merck Sequant as mobile phase and were detected by UV toine, concentration and peak areas showed good linear relationships between 4.7 - 470 mg/L with R^2 = 0. 9999. Inter-day RSD was 0. 36% and 0. 42%, intra-day RSD was 1. 1% and 2.4% , the limit of detection was 0. 061 mg/L and 0. 065 mg/L, and the limit of quantitation was 0. 23 mg/L and 0.25 mg/L respectively. Average recovery was between 89. 6% - 100. 7%. The method showed simplicity, good repeatability and high sensitivity.
