摘要:阿卡波糖(1)是游动放线菌(Actinoplanes sp.)的次级代谢产物,其生成与菌体生长具有相关性。发酵液渗透压对细胞生长及1的生物合成影响显著。本研究考察了渗透压对1生物合成的影响,结果表明通过补料将发酵前期渗透压维持在300mOsm/kg,中后期维持400~500mOsm/kg,麦芽糖浓度维持6%,1生物合成水平大幅提高,50L发酵罐效价为3360mg/L,提高了50%以上。
Acarbose biosynthesis showed a growth-associated type of product formation in Actinoplanes sp. fermentation. It was found that medium osmotic pressure affected both cell growth and acarbose biosynthesis notably. A significant increase of acarbose production was achieved when osmotic pressure was kept at 300 mOsm/kg in the basal medium and at 400 - 500 mOsm/kg during the fermentation process as well as the maltose concentration was kept about 6%. The titer of acarbose reached to 3 360 mg/L in a 50 L fermentor, which was increased by 50%.
