摘要:本文分析鸟苷产生菌枯草芽孢杆菌 (Bacilussubtilis)在 5 0L多参数自控发酵罐上的发酵过程特点 ,基于多种在线及离线参数的检测 ,通过相关分析将生理调控的工艺参数和生物合成过程中的代谢流分布相联系 ,发现了发酵过程中的代谢流向糖酵解和TCA循环的迁移 ,并初步分析了产生代谢流迁移的原因 ,在此基础上优化发酵过程使产苷水平稳定在 3 0g L。
The characteristic of Bacillus.subtilis fermentation process of guanosine on 50L fermentor was analyzed. Based on determination of on-line and off-line parameter, using correlation analysis, the technology study of physiologic regulation was combined with the metabolic flux distribution of synthesis process. The metabolic flux shift from HMP to EMP and TCA cycle during fermentation was found. The reason of the flux shift was preliminary analyzed, based on which the procedure was optimized to increase the yield of guanosine to 30g/L.
