摘要:NOVELTY - Constructing Escherichia coli strain capable of exhibiting production of succinic acid, involves performing transformation of pathway by blocking tricarboxylic acid (TCA) cycle, and/or enhancing acetic acid uptake and oxaloacetate supply, deleting by-product formation pathway, and/or reducing ineffective circulation caused by producing acetic acid from pyruvic acid through decarboxylation, and/or dredging acetyl coenzyme (CoA) node metabolic flux, deleting lactic acid and ethanol production pathway key genes to ease acetyl CoA node metabolic flux. USE - The method is useful for constructing E.coli strain useful for producing succinic acid (all claimed). ADVANTAGE - The method provides E.coli strain that produces succinic acid with no by-product. DETAILED DESCRIPTION - Method for constructing Escherichia coli strain capable of exhibiting production of succinic acid by metabolic engineering, where metabolic engineered E.coli utilizes acetic acid as raw material for succinic acid fermentation, involves performing transformation of pathway by blocking tricarboxylic acid (TCA) cycle, and/or blocking succinic acid utilization pathway, and/or enhancing acetic acid uptake and oxaloacetate supply, strengthening glyoxylate cycle, and/or deleting by-product formation pathway, and/or reducing ineffective circulation caused by the producing acetic acid from pyruvic acid through decarboxylation, and/or dredging acetyl coenzyme (CoA) node metabolic flux, and/or deleting lactic acid and ethanol production pathway key genes to ease acetyl CoA node metabolic flux. INDEPENDENT CLAIMS are included for the following: (1) E.coli strain obtained by metabolic engineering; and (2) method for producing succinic acid using the E.coli strain.
