摘要:研究了在复合培养基中发酵生产必特螺旋霉素时,铵离子浓度对发酵组分的影响。从发酵24 h开始,每12 h补加3.2 mmol/L的铵离子,考察其对必特螺旋霉素中异戊酰螺旋霉素组分和异戊酰基螺旋霉素Ⅲ/Ⅱ的调节作用。实验结果表明:在降低基础培养基铵离子浓度的基础上,补加铵离子可以明显改善必特螺旋霉素组分,生物效价达2 200 u/mL。通过测定发酵过程中还原糖、相关有机酸、琥珀酸脱氢酶(Succinate dehydrogenase,SDH)活性、缬氨酸脱氢酶(Valine dehy-drogenase,VDH)活性等中间代谢数据,对铵离子的作用原理进行了讨论。
In previous research it was proved that low ammonium ion concentration in the complex media had negative effect on the titer of bitespiramycin fermentation though percentage of isovalerylspiramycin was greatly enhanced. In present study, feeding of ammonium ion during fermentation was found to increase the growth and titre of bitespiramycin as well as the percentage of isovalerylspiramycin and the ratio of isovalerylspiramycin Ⅲ to isovalerylspiramycin Ⅱ. Based on the data of fermentation process, such as residual sugar, organic acids, the activities of succinate dehydrogqenase and valine dehydrogenase, the mechanism of modulating growth by ammonium ion and metabolism of Streptornyces spirarnyceticus were discussed.
