摘要:以肝素黄杆菌为研究对象,提出了一种将发酵分为生长和产酶两个阶段的生产肝素酶的新工艺。对培养基组成进行了优化,优化后组成(g/L)葡萄糖8,氯化铵4,磷酸二氢盐6,组氨酸0.75,甲硫氨酸0.75,氯化镁0.5;微量盐10-4mol/L。菌体生长和产酶两个阶段的最优温度为27°C和23°C,pH为6.4和7.0。在诱导时加入10-4mol/LBa2+可以较好地消除SO2-4的阻遏作用。采用优化工艺,摇瓶产酶比原来提高了66%。
A novel fermentation processes to produce heparinase by Flavobacterium heparinum were established. The fermentation is divided into two stages: bacteria growth and enzyme production. The optimized culture medium composition were as follows(g/L), glucose 8, NH4Cl 4, NaH2PO4 3, KH2PO4 3,histidine 0.75, methinine 0.75, MgCl2 0.5 and trace elements 10^-4 mol/L. The optimum temperatures and pH for bacteria growth and enzyme production were 27 ℃ and 23 ℃, and 6.4 and 7.0 respectively. Addition of Ba^2+ at 10^-4 mol/L could get rid of sulfur repression which led to the increase of the total enzyme productivity of heparinase by 66 %.
