摘要:目的:采用反相高效液相色谱-二极管阵列检测器(RP-HPLC-DAD)和高分辨电喷雾飞行时间质谱(ESI-TOF-MS)联用技术,对生大黄及其炮制品中的主要成分进行鉴定,并考察炮制过程对大黄有效组分的影响。方法:采用HPLC/UV/MS对生大黄和制大黄的小分子化合物进行定性鉴定;采用比色法测定生、制大黄中鞣质及多糖的含量;采用高效液相色谱法测定生、制大黄中没食子酸及游离蒽醌的含量。结果:HPLC/UV/MS分析表明,大黄中主要含有多酚类、蒽醌衍生物类等成分;大黄经炮制后,各组分含量变化各不相同。
AIM : To investigate the constituents of Radix et Rhizoma Rhei and processed Radix et Rhizoma Rhei by HPLC-electrospray-time of flight mass spectrometry ( ESI-TOF-MS), and compare the difference between the chemical materials. METHODS : The soluble components of Radix et Rhizoma Rhei and processed Radix et Rhizoma Rhei were analyzed by HPLC/UV/MS technique. The tannin and polysaccharide contents of Radix et Rhizoma Rhei and processed Radix et Rhizoma Rhei were determined by colorimetry, while the contents of gallic acid and free anthraquinone of Radix et Rhizoma Rhei and processed Radix et Rhizoma Rhei were analyzed by HPLC technique. RESULTS: Phenols and anthraquinones are the major components of Radix et Rhizoma Rhei. Compared with raw Radix et Rhizoma Rhei, the contents of sereral compounds of processed Radix et Rhizoma Rhei were changed.
