摘要:采用单因素和响应面实验优化了高温高压-复合酶法处理香菇(Lentinula edodes)废弃菌糠的条件:料液比1∶20(g∶mL),最佳复合酶配比为纤维素酶∶木聚糖酶∶β-葡聚糖酶=3∶2∶1,总加酶量660U·g^(-1),pH 5.20,温度45℃,酶解时间6h,在此条件下总糖提取率为13.4%,提取物得率为22.4%。体外抗氧化实验表明,高温高压-复合酶法降解香菇菌糠得到的提取物(LR-UE)较水提法和高温高压法得到的提取物(LR-W和LR-U)具有更强的DPPH和ABTS自由基清除能力。体外细胞实验表明,LR-UE能提高脂多糖(LPS)刺激巨噬细胞RAW 264.7释放NO的抑制率,表现出良好的体外抗炎活性。研究结果为香菇菌糠的高值化利用提供了新思路。
The rapid development of Lentinula edodes cultivation in China has brought about a substantial increase in cultivation residue waste which is calling for sustainable and eco-friendly recycling.In this study,single factor and response surface experiments were carried out to optimize the conditions for processing L.edodes cultivation residue by high temperature and pressure-complex enzyme.The optimal conditions were as follows:complex enzyme composed of cellulase,xylanase andβ-glucanase at 3∶2∶1,complex enzyme quantity of 660U·g^(-1),material-liquid ratio of 1∶20(g∶mL),pH 5.20,temperature at 45℃,enzymolysis time for 6h.Using these conditions,the overall extraction rate was 22.4%,and the total sugar extraction rate was 13.4%.Antioxidant experiments in vitro showed that the extract obtained through the high temperature and pressure complex enzyme(LR-UE)had stronger activities on scavenging 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH)and 2,2′-diazo-bis-3-ethylbenzothiazolin-6-sulfonic acid(ABTS)than the extracts obtained by water extraction(LR-W)and high temperature and pressure(LR-U).LR-UE also increased the inhibition rate of NO release from RAW264.7macrophage cells upon lipopolysaccharide(LPS)stimulation,showing good anti-inflammatory activity in vitro.These results provided a new idea for highvalue utilization of Lentinula edodes cultivation residue.
