摘要:用异丙基 -β-D-硫代半乳糖苷 (IPTG)、乳糖、巯基乙醇对重组人锰超氧化物歧化酶(rh Mn-SOD)工程菌进行诱导 ,结果表明 3种物质都可以诱导 rh Mn-SOD外源蛋白的表述。通过酶活力、比活测定及电泳的结果显示 ,以乳糖作为诱导剂的表达效果明显好于 IPTG和巯基乙醇 ,进一步的实验结果表明乳糖诱导的最佳浓度为 80
Three kinds of materials including IPTG, lactose, and mercaptoetanol were used as the inducers to induce the expression of recombinant human manganese superoxide dismutase. The results show that all these materials can induce the expression of exogenous rhMn SOD protein. By assaying the enzyme activity, specific activity and applying electrophoresis, the results show that the induction effect of lactose is better than that of the other two materials. And further experimental result indicate that the optimum concentration of lactose is 80mmol/L.
