摘要：综述治疗高脂血症的他汀类药物阿托伐他汀钙的合成方法,根据文献报道归纳出阿托伐他汀钙的合成路线有两种,一种是先合成出取代的吡咯环,然后在环上引入手性的3,5-顺式双羟基庚酸结构(线性合成),一种是先制备手性的3,5-顺式二羟基庚酸片断,然后与1,4-二羰基化合物环合得到吡咯环结构(汇聚合成)。还比较了各合成工艺方法的优缺点,为寻找一条适合大规模、工业化生产的工艺路线提供参考。
This review summarized the various synthetic processes of atorvastatin calcium.There are two ways of synthesis of atorvastatin calcium based on the references.One is to synthesize the substitued pyrrole ring firstly,the introduct chiral 3,5-cis-dihydroxy heptanoic acid structure into the ring（linear synthesis）.The other one is to prepare chiral 3,5-cis-dihydroxy heptanoic acid fragment,the form the pyrrole ring structure with 1,4-dicarbonyl compounds（pool synthesis）.By comparison of the merits and demerits of the process for the preparation of atorvastatin,try to find a route which shows more favorable commercial value in the manufacturing scale.
