摘要:整个测量系统由沈阳电影反光镜广制造的LZ-1000型组合式流动注射分析(FIA)仪与固定化青霉素酶反应器组成。测定原理同碘量滴定测量青霉素,实验表明:线性范围在0.06~0.75mmol/L(取样40μl),线性相关系数0.998。重现性:对0.12、0.75mmol/L浓度的青霉素反复测定8次,平均值分别为0.690mv、3.838mv,标准差分别为0.0066mv、0.0164mv,精密度分别0.82%、0.34%。测定条件影响:在青霉素浓度低于2·5mmol/L时,磷酸盐浓度在0.05~0.25mol/L,pH在6.0~7.5,测定温度在8℃~30℃对测定结果几乎没有影响。测定速率可达每小时120样。在室温(8~20℃)下使用,每天测定400样,使用30天未见活力下降。4℃下保存近半年未见酶反应器活力下降。
An automated assay for the
determination of penicillin in broth is presented. The method was based on the classical
iodometric penicillin assay which is incorporated in a flow injection analysis(FLA)system.The
required hydrolysis was performed on line by using an immobilised penicillinase reactor.
Various factoers which influence the response of this system and the performance of the
system were descri-bed.Good linear relationship was obtained within the ran9of 0.06~0.75
mmol/L(40μL)penicillin. The correlation coefficient of the calibration curve is 0.998.The speed
of determination of this system is high(60~120samples per hour).The enzyme in the reactor is
stable for at least 6mo-nths.Recycle life of the reactor is more than 30 days.
