摘要:为探索新近暴发的甲型H1N1流感病毒各基因片段的变异性与本次疫病流行的内在原因,以NCBI GenBank中报道的1株新发的典型H1N1甲型流感病毒A/New York/1669/2009(H1N1)作为研究对象,运用在线BLAST和CLUSTAL X等生物信息学软件对其PA、NP、M、PB1、PB2、NA、HA和NS基因片段进行了比较分析。结果表明,新发甲型H1N1流感毒株的PB1、NP基因来源于人源谱系流感病毒,HA、NA、NS、M基因来源于猪源谱系流感病毒,PB2和PA基因来源于禽源谱系流感病毒。此外,新发甲型H1N1流感病毒的HA和NA分别在第249和386位新出现1个"-NTT-"、"-NFSI"-的糖基化位点。HA的6个潜在抗原决定簇中有3个发生了氨基酸序列改变,NA则是12个中有5个发生了改变。表明,这些抗原基因的大范围重组与高频率变异可能是造成这次流感暴发与大规模流行的根本原因。
In order to explore the internus reasons of the recent pandemic influenza and the variability of eight gene fragments of influenza A(H1N1) virus,a typical influenza A(H1N1) virus A/NewYork/1669/2009(H1N1) broke out recently was analyzed and compared on its eight gene fragments of PA,NP,M,PB1,PB2,NA,HA,and NS by BLAST and CLUSTAL X bioinformatics softwares.The results showed that PB1 and NP genes derived from human influenza virus,and HA,NA,NS,and M genes derived from swine influenza virus,while PB2 and PA genes derived from avian influenza virus.A new glycosylation site-NTT-was found in position site 249 of HA gene,and another new glycosylation site-NFSI-was found in position site 386 of NA gene,too.Meanwhile,the changes of 3 amino acids on HA gene led to the changes of 6 potential antigenic determinants,and 5 of 12 potential antigenic determinants of NA gene changed.The recombination and high-frequency variability of these antigenic determinants might be the primary cause of the pandemic influenza outbreak.
