摘要:在 2 .6L全自动发酵罐中对人 α2 a干扰素重组酵母的补料分批培养过程作了初步研究。结果表明 ,培养过程中待葡萄糖耗尽时流加葡萄糖 ,并维持低糖浓度 ;在培养 1 0 h~ 2 0 h时 ,恒速流加 2 0 mg/L的腺嘌呤 ,能较大幅度提高表达水平 ,生物活性从原工艺的 3.1× 1 0 9IU/L提高至1 .3× 1 0 10 IU/L。表达期 p H值对生产水平有很大影响 ,宜控制在 5.0~ 5.
Production of recombinant human interferon α2a by Saccharomyces cerevisiae in fed batch culture was investigated systematically in 2.6 L automatic jar fermenter. During the fermentation, suitable amount of glucose was added to keep the glucose concentration at low level, when glucose was exhausted; 20mg/L of adenine was supplemented at even speed during 10h~20h of fermentation. As a consequence, the expression level of human interferon α2a has increased by 4 times to 1.3×10 10 IU/L. In expression phase, pH value has an important impact on expression level, which should be controlled within 5.0~5.3.
