摘要:通过5-甲基-5-苄氧羰基-三亚甲基碳酸酯(MBC)和L-丙交酯(LLA)开环共聚,合成了完全生物可降解的乳酸——功能化碳酸酯的共聚物(PLM).PLM侧链苄基在Pa(OH)2、H2条件下脱除后,得到侧链含有羧基的聚合物(d-PLM).d-PLM膜表面经1-乙基-(3-二甲基氨基丙基)碳酰二亚胺盐酸盐(WSC)活化后接枝肝素分子,得到肝素化乳酸——碳酸酯共聚物(Hep-PLM).经测定,Hep-PLM膜表面肝素含量达到6.33μg/cm2,亲水性较聚左旋丙交酯(PLLA)有明显提高.支架体外细胞培养试验显示,Hep-PLM具有良好的生物相容性.动物实验结果表明,Hep-PLM作为支架涂层材料对血管壁的炎性刺激明显低于PLLA,有望成为一种理想的生物可降解支架涂层材料.
Biodegradable polymer including L-lactide (LLA) and 5-methyl-5-benzyloxycarbonate-l,3-diox- an-2 -one (MBC) was synthesized by ring-opening polymerization. The polymer was deprotected under hy: drogen atmosphere using Pd(OH)2/C as a catalyst. Heparin was conjugated to the surface of the deprotected polymer film activated by WSC. The surface exposed heparin content associated was measured to be 6.33μg/cm2. The cell culture results show good biocompatibility of the heparin immobilized material and this novel polymer can be applied as a blood biocompatible and biodegradable coating material for coronary stents with lower inflammation formation and higher hydrophilicity than polylactide. This strategy may reduce implant-associated adverse responses and be a better approach for vascular tissue engineering applications.
