摘要:基于Gabriel反应,在萘酰亚胺的4-位引入邻苯二甲酰亚胺、亚胺位引入溶酶体定位基团吗啉,合成了探针NMA和NMB用于对肼的检测。实验结果表明两种探针对肼均有较强的光谱响应,其中NMA对肼的响应更加灵敏、快速。肼的加入使NMA的吸收光谱红移100 nm,其在527 nm处的荧光信号最大增强约500倍。在5.0~70μmol/L范围内,NMA的荧光强度与肼的浓度呈现良好的线性关系,对肼的检出限为1.83μmol/L。此外,通过高效液相色谱和密度泛函理论计算验证了探针NMA对肼的响应机理。细胞成像结果表明NMA能够实现对溶酶体内肼的荧光成像。
Based on the Gabriel reaction,two fluorescent probes,NMA and NMB,were synthesized for hydrazine detection.The probes were constructed with naphthalimide as the fluorophore,a phthalimide at the 4-position selected as the receptor and a morpholine at imide site as a lysosome-targeted group.Experimental results demonstrated that both probes exhibited evidently spectral responses to hydrazine.Compared to NMB,NMA displayed better performance such as higher sensitivity and faster response.The addition of hydrazine resulted in~100 nm red shift in the absorption of NMA,companied with 500-fold fluorescence enhancement.The fluorescence intensity of NMA showed a good linear relationship with hydrazine concentration in the range of 5.0-70μmol/L,with a detection limit(3σ/k)of 1.83μmol/L.The response mechanism of NMA to hydrazine was validated by HPLC and theoretical calculations.Confocal imaging experiments demonstrated that NMA could realize imaging lysosomal hydrazine.
