摘要:利用脱氮假单胞菌发酵生产维生素B12的过程中,甜菜碱是产物合成的前体,但高浓度的甜菜碱对细胞生长会产生抑制作用,将发酵液中甜菜碱浓度控制在适宜水平对维生素B12的高效生产有着重要的意义。考察了不同补料策略下,不同的甜菜碱浓度对维生素B12合成的影响。实验结果表明:在连续发酵过程中,维持低浓度的甜菜碱有利于维生素B12向胞外释放,尤其在产物大量合成期,低浓度甜菜碱能够明显促进胞外产物的积累。在50 L发酵罐中,当甜菜碱质量浓度控制在3.0 g/L时,维生素B12的产量最高,是甜菜碱质量浓度为9.0 g/L时的129%。
Although betaine is one of the crucial precursors in vitamin B12(VB12) fermentation by Pseudomonas denitrifican,the cell growth will be inhibited at its high concentration.So it is important to keep the betaine concentration under suitable level for high VB12 production.The effects of betaine of different concentrations on the VB12 production were investigated with different feeding strategies.The results indicated that the extracellular release of VB12 could be stimulated by maintaining a low betaine concentration,and it also showed that the low betaine concentration could significantly enhance the accumulation of VB12 especially in the late fermentation.The highest VB12 production could be achieved when the betaine concentration was kept at 3.0 g/L in the 50 L fermenter,which was 129% of that at 9.0 g/L.
