摘要:目的:探讨高压氧(HBO)对荷Lewis肺癌小鼠在化学治疗中的增效作用及其机理。方法:270只荷Lewis肺癌的C57小鼠被随机等分为空气对照组、高压氮氧组、HBO组、HBO-丝裂霉素组、HBO-环磷酰胺组、HBO-SOD组、丝裂霉素组、环磷酰胺组和SOD组共9个组。对Lewis肺癌小鼠使用化疗结合HBO疗法。采用电子自旋共振(ESR)技术直接检测Lewis肺癌组织中的氧自由基(OFR)含量,并且应用分光光度法测定其脂质过氧化物丙二醛(MDA)的含量,同时对各组动物荷瘤的体积、重量、坏死率及小鼠致死率进行了观察。结果:HBO组、HBO-丝裂霉素组及HBO-环磷酰胺组的动物与相应的对照组相比,肺癌组织中的OFR含量和MDA含量均显著增高(P<0.05~0.001);荷瘤组织坏死率高,且坏死所需时间短;动物成活率高;肿瘤体积和重量也显著小于对照组(P<0.05~0.001)。结论:荷Lewis肺癌小鼠经HBO暴露可以减缓荷瘤生长速度并加速癌细胞的坏死,丝裂霉素或环磷酰胺结合HBO治疗能显著增强药物的抗肿瘤作用,减轻药物的副作用和延长小鼠的生命。
Aim:To study the synergetic effect and its mechanism of hyperbaric oxygen(HBO) and chemo therapy on Lewis lung cancer in mice.Methods:270 C 57 mice with Lewis lung cancer were randomly divided into the air control group,normoxic hyperbaric nitrox group,HBO group,HBO SOD group,HBO mitomycinum group,HBO cyclophosphamidum group,mitomycinum group,cyclophosphamidum group and SOD group.The mice with the Lewis lung cancer were treated with combined chemo therapy and HBO.The contents of oxygen free radicals(OFR) in Lewis lung cancer tissue were measured by eletron spin resonance(ESR) technique and that of malondialdehyde(MDA) by spectrophotometry;at the same time the volume,weight,necrosis of tissue and fatality of mice in different groups were observed.Results:The contents of OFR and MDA in tumor tissue in the HBO group,HBO mitomycinum group and HBO cyclophosphamidum group were markedly higher than those in control groups( P <0 05 ̄0 001).The necrosis rate of tumor tissue and the survival rate of mice were higher,the necrosis time was shorter,and the volume and weight of tumor tissue were obviously smaller or lighter than those in control groups( P <0 05 ̄0 001).Conclusion:The results suggest that the HBO exposure may slow the growth and hasten the necrosis rate of Lewis lung cancer cells in mice.The therapeutic effect of mitomycinum or cyclophosphamidum on the tumor could be enhanced by HBO,meanwhile the side effects of anti tumor drugs mitigated and the lives of mice prolonged.
