摘要:采用本实验室筛选到的酿酒酵母LH1催化苯甲酰甲酸甲酯不对称合成(R)-(-)-扁桃酸甲酯。在单相体系中考察了初始底物浓度、细胞浓度、pH、温度、辅助底物、葡萄糖浓度和反应时间等因素对产物得率和对映体选择性的影响,获得了较佳的还原条件。当细胞浓度75 g(干细胞)/L,葡萄糖浓度30 g/L,底物浓度100 mmol/L,pH 8.0,温度30℃,反应时间30 h时,产物扁桃酸甲酯的得率达94.3%,(R)-(-)-扁桃酸甲酯的对映体过量值(ee)达95.0%。在苯甲酰甲酸甲酯的转化反应中,酿酒酵母LH1菌株表现出非常好的对映体选择性,且ee值受环境因素的影响非常小。
(R)-(-)-Methyl mandelate was produced by asymmetric reduction of methyl benzoylformate catalyzed by Saccharomyces cerevisiae LH1 screcned by our laboratory. The effects of some factors including initial substrate concentration, amount of yeast cells, pH, temperature, co-substrate as well as the reaction time on the yield and enatiomeric excess (ee) of product in mono-phase system were examined. The optimal reaction conditions are as fallows: cell concentration 75 g(DCW)/L, glucose concentration 30 g/L, substrate concentration 100 retool/L, pH 8.0, temperature 30 ℃, reaction time 30 h, and the yield 94.3% with the high ee of 95%. In the bioreduction of methyl benzoylformate, S. cerevisiae LH1 exhibited high enantioselectivity and ee was little affected by environmental factors.
