摘要:采用乳糖作为诱导物 ,对1 0启动子控制下重组蛋白 rh NTA ( a new thrombolyticagent)在大肠杆菌中的表达进行研究 ,结果为培养 1 2 h(诱导 6h)后湿菌体重量 86.0~ 1 0 0 .2 g/L、目标蛋白占细胞总蛋白的 40 %~ 50 %。研究了关键基质组分、乳糖诱导等对表达的作用。结果表明 ,基础料和补充氮源中酵母膏的比例、诱导时机和乳糖流加方式等对表达量的提高有明显的影响。实验显示 。
The process of expression of rhNTA (a new thromobolytic agent) gene using lactose as an inducer under the control of φ10 promoter in Escherichia coli is investigated. The recombinant protein rhNTA is 40%~50% of the total cell protein and the wet cell weight is 86.0~100.2 g/L of the culture broth after 12 h cultivation. It is manifested that the key components of the subtract and the strategy of lactose induction play an important role with regard to expression of recombinant protein. The percentage of yeast extract of the nitrogen source during the growth phase and in the basal medium during the phase of induction and the time and condition that the induction starts are effective. The target protein in the cell of Escherichia coli was mainly occupied by soluble part of rhNTA protein.
