摘要:在用林可链霉菌发酵生产林可霉素的过程中,采用FUS-50L多参数全自动发酵罐,通过在线参数、离线参数等的关联分析,发现中后期的调控非常关键。从80h开始补入适量的玉米浆和碳酸钙,可增加三羧酸循环的通量,强化菌体的维持代谢和次级代谢,184h时,发酵液的生物效价由4792μg/mL(原工艺)增加到7543μg/mL(优化工艺);同时,结合生产实际,将50L罐的优化实验结果应用到60t生产罐实验,184h时,发酵液的生物效价由4536μg/mL(原工艺)增加到6582μg/mL(优化工艺)。有效解决了生产后期生物效价增幅很小的问题。
Through correlative analysis among the online and off-line parameters and so on, it was found that regulation and control was crucial in the mid-late stage during mid phase in the course of fermentation for producing lincomycin by Streptomyces lincolnensis in the FUS-50L automatic multi-parameter fermentor. Begining from 80 hours, after adding proper amount corn steep liquor and CaCO3 to the fermented broth, the tricarboxylic acid circulation flux increased, maintenance metabolism and secondary metabolism intensity increased too, When 184h, lincomycin production increased from 4792μg/mL (original production process) to 7583μg/mL (optimized production process) ; At the same time, considering the practical situation of production, the optimized experimental result of FUS-50L fermentor was applied to the production experiment in the 60 ton fermentor, When 184h, the titres of the fermented broth increased from 4536 μg/mL (original production process) to 6582μg/mL (optimized production process). The question that the increased scope of titres was very small during later period was solved effectively.
